Transparent Embryos

Transparent embryos are embryos that allow light to pass through their tissues, either because they naturally lack substantial pigmentation or because researchers reduce optical barriers, making internal development visible. Their transparency enables microscopy to track cells, tissues, and organ formation in living specimens, often using fluorescent labels that reveal specific structures as they change over time. In biology, this approach supports direct study of embryonic development, cell movement, gene expression, and disease-related abnormalities while reducing the need to disrupt samples. Transparent embryo models also help connect cellular mechanisms with whole-organism outcomes and improve the visualization of developmental processes.

We describe a protocol for xenotransplantation into the yolk of transparent zebrafish embryos that is optimized by a simple, rapid staging method. Post-injection analyses include survival and assessing the disease burden of xenotransplanted cells by flow cytometry.

Microinjection of Live Drosophila Embryos: Early Delivery of Reagents to the Developing Embryo

2023

This video describes how to perform microinjections that deliver reagents into live Drosophila embryos. Researchers can use this method to introduce exogenous compounds like nucleic acids and proteins during the early stages of embryo development. The featured protocol demonstrates how to set up and perform the procedure for injection of any soluble reagent–here, a fluorescent protein for live imaging experiments.

Zebrafish Breeding and Embryo Handling

2023

Zebrafish (Danio rerio) are an important model organism that is particularly valuable for research in developmental biology. Zebrafish are extremely fertile and can produce hundreds of progeny per week, so it is relatively easy to collect a large number of embryos for high sample numbers. Furthermore, zebrafish undergo rapid development and embryos are transparent, allowing for easy visualization of developmental processes. This video covers the steps required for the collection of newly...

The Mammalian Ocular Lens in Focus: Development, Anatomy, Physiology, Transparency, Biomechanics, and Age-Related Challenges

We describe the experimental method to deposit nanostructured oxide thin films by nanosecond Pulsed Laser Deposition (PLD) in the presence of a background gas. By using this method Al-doped ZnO (AZO) films, from compact to hierarchically structured as nano-tree forests, can be deposited.
