Viral Uptake

Viral uptake is the process by which virus particles enter host cells, a critical step in infection and in the delivery of viral genetic material. It typically begins when viral surface proteins bind specific receptors on the cell membrane, triggering membrane fusion or receptor-mediated endocytosis; subsequent uncoating releases the viral genome into the cell. Studying viral uptake helps explain host range, tissue tropism, and infection mechanisms, while supporting the development of antiviral therapies, vaccines, and viral vectors for gene delivery. Experimental analysis of this process can also reveal cellular barriers that limit or promote viral entry.

Source: Cafini, F., et.al. Methodology for the Study of Horizontal Gene Transfer in Staphylococcus aureus. J. Vis. Exp. (2017)This video demonstrates competence-induced DNA uptake in Staphylococcus aureus through natural transformation. A chloramphenicol-sensitive strain engineered to constitutively express a sigma factor is cultured under nutrient-limiting conditions to enhance competence, then incubated with donor DNA carrying a resistance gene. Successful gene transfer is confirmed by the...
