Cell Culture Model

A cell culture model is an experimental system in which living cells are maintained and studied outside an organism under controlled laboratory conditions, allowing researchers to examine biological processes in a simplified setting. Cells are grown in nutrient-containing media and provided with suitable temperature, gas exchange, and surface or matrix conditions; their behavior depends on cell type, density, and signals from the surrounding environment. In biology, these models support investigations of cell growth, differentiation, communication, and responses to disease-related or chemical stimuli. They also provide tractable platforms for comparing mechanisms, evaluating potential treatments, and guiding more complex studies in tissues or whole organisms.

Traditional, two dimensional cell culture techniques often result in altered characteristics with respect to differentiation markers, cytokines and growth factors. Three-dimensional cell culture in the rotating cell culture system (RCCS) reestablishes expression of many of these factors as shown here with an extravillous trophoblast cell line.

A cell culture model of resistance arteries is described, allowing for the dissection of signaling pathways in endothelium, smooth muscle, or between endothelium and smooth muscle (the myoendothelial junction). The selective application of agonists or protein isolation, electron microscopy, or immunofluorescence can be utilized using this cell culture model.

This video demonstrates a method to establish a three-dimensional tumor spheroid and T cell co-culture. An agarose cast with microwells containing tumor spheroids is prepared, followed by the addition of T cells. After incubation, this setup allows studying the interaction of T cells with tumor spheroids in a controlled three-dimensional environment.

Organotypic hippocampal slice cultures (OHSC) represent an in vitro model that simulates the in vivo situation very well. Here we describe a vibratome-based improved slicing protocol to obtain high quality slices for use in assessing the neuroprotective potential of novel substances or the biological behavior of tumor cells.
