Lentiviral Particle Production

Lentiviral particle production is a laboratory process that generates replication-incompetent viral vectors for delivering genetic material into target cells. It typically involves introducing vector, packaging, and envelope components into producer cells, which assemble and release lentiviral particles that can enter recipient cells and integrate the carried transgene into the host genome. Researchers use these vectors to achieve stable gene expression in cultured cells and model systems, supporting studies of gene function, disease mechanisms, and cell-based therapies. Careful control of production and characterization helps improve vector quality, delivery efficiency, and experimental reproducibility.

We describe the production strategy of integrase-deficient lentiviral vectors (IDLVs) as vehicles for delivering CRISPR/Cas9 to cells. With an ability to mediate quick and robust gene editing in cells, IDLVs present a safer and equally effective vector platform for gene delivery compared to integrase-competent vectors.
