Organoid Media Preparation

Organoid media preparation is the controlled formulation of culture medium used to establish, maintain, and study three-dimensional organoids, which model tissue organization in the laboratory. It combines a basal medium with defined nutrients, signaling molecules, and other supplements that support cell survival, self-organization, and proliferation under sterile culture conditions; adjusting these components can influence organoid growth and phenotype. In cancer research, carefully prepared media help sustain patient-derived and tumor organoids for investigating cancer biology, evaluating drug responses, and modeling treatment resistance. Standardized preparation improves reproducibility, enabling comparisons across experiments and supporting more physiologically relevant studies than conventional two-dimensional cultures.

Preparing Organoids from Tumors: A 3D In Vitro Tumor Model

2023

Organoids derived from tumors are a type of 3D cell culture model that can maintain the pathological features and organ-specific cell lineage of tumors. In the example protocol, we will see scientists generate organoids from a prostate tumor from a genetically engineered mouse model.

This video describes the electroporation protocol for transfecting large plasmids into three-dimensional gastrointestinal organoids to prove its universal functionality in different entities, namely pancreatic ductal adenocarcinoma (PDAC), colorectal cancer (CRC), cholangiocarcinoma (CCC), and gastric cancer (GC) organoids.

Generating an Organoid Culture from a Brain Tumor Cell Suspension

2025

This video describes the preparation and cultivation of brain tumor organoids from single-cell suspension. Using a laminin-rich extracellular matrix as a scaffold and culture media for nutrients, cells are facilitated to multiply and self-organize within the organoid. Consequently, ex vivo organoids are generated, mimicking cellular diversity and the microenvironment of brain tumors.

An In Vitro Method to Generate Human Brain Organoids

2025

The video demonstrates a method for generating human brain organoids from induced pluripotent stem cells. It shows how the neurosphere forms a neuroectodermal layer, which suppresses the formation of other germ layers. The neurospheres are embedded in a 3D gel matrix and form neuroepithelial buds. When incubated with organoid media, these neuroepithelial buds differentiate into specific brain regions, resulting in the formation of mature brain organoids.
