Carbene Precursor Isolation

Carbene precursor isolation is the preparation, purification, and storage of compounds that can generate highly reactive carbenes under controlled conditions. These precursors, which may include diazo compounds or related masked carbene sources, are isolated by limiting heat, light, moisture, and catalytic impurities that could trigger nitrogen loss or other decomposition pathways. Careful handling and characterization establish precursor identity, purity, and stability before use. Isolated carbene precursors support controlled cyclopropanation, insertion, rearrangement, and metal-carbene reactions, allowing researchers to study reactive intermediates while improving reaction reproducibility and laboratory safety.

Isolating Free Carbenes, their Mixed Dimers and Organic Radicals

We present protocols for the isolation of stable heterocyclic carbenes. The synthesis of a cyclic (alkyl)(amino) carbene (CAAC) and an N-heterocyclic carbene (NHC) is demonstrated using filter cannulas and Schlenk technique. We furthermore present the synthesis of the related oxygen-sensitive, electron-rich mixed “Wanzlick dimer” and the reduced stable organic radical.

Here we describe a protocol for isolating subsets of precursor B-cells from umbilical cord blood. A sufficient quantity and quality of nucleic acids may be extracted from the cells and used in subsequent assays utilizing DNA or RNA.

This isolation method allows simultaneous purification of three multipotent precursor cell populations respectively from three structural layers of blood vessels: myogenic endothelial cells from intima, pericytes from media, and adventitial cells from adventitia.
