Simulated Gastrointestinal Media

Simulated gastrointestinal media are laboratory-prepared chemical solutions that reproduce the conditions of the stomach and intestines to study how substances behave during digestion. Their composition can be adjusted to reflect factors such as pH, ionic strength, digestive enzymes, bile components, and sequential movement from gastric to intestinal environments, allowing researchers to model chemical transformations and dissolution processes under controlled conditions. In chemistry, these media support evaluations of drug release, nutrient digestion, solubility, stability, and bioaccessibility without relying solely on in vivo experiments. Standardized formulations also improve comparisons among formulations and help guide pharmaceutical, food, and materials research.

A pendant drop surface film balance implemented with a multi-subphase exchange, nicknamed the OCTOPUS, allows for mimicking digestive conditions by the sequential subphase exchange of the original bulk solution with simulated gastrointestinal fluids. The simulated in vitro digestion is monitored by recording in situ the interfacial tension of the digested interfacial layer.

Here, a protocol to isolate and establish primary fibroblast/myofibroblast (MF) cultures from frozen gastric, small intestinal, and colonic tissue-yielding cells with a MF phenotype-is presented. These cells express CD90, α-SMA and vimentin. MFs can be used for a variety of functional assays including enzymatic activity and cytokine production.

This protocol describes the use of excised intestinal tissue preparations or "intestinal sacs" as an ex vivo model of intestinal barrier function. This model may be used to assess integrity of both the epithelial barrier and the mucous gel layer at specific intestinal sites in animal models of digestive disease.
