Acute Hyperglycemia Model

An acute hyperglycemia model is an experimental system that produces a short-term elevation in glucose to examine how developing organisms or tissues respond to excess sugar. In developmental biology, researchers expose embryos, cells, or tissues to defined high-glucose conditions for a controlled period, then assess changes in development, cell behavior, and molecular signaling. This model helps distinguish immediate effects of hyperglycemia from those caused by prolonged metabolic disease and can reveal links between abnormal glucose exposure and developmental outcomes. Its controlled design supports studies of embryonic growth, diabetes-associated developmental complications, and potential mechanisms underlying glucose-sensitive defects.

Acute and Chronic Models of Hyperglycemia in Zebrafish: A Method to Assess the Impact of Hyperglycemia on Neurogenesis and the Biodistribution of Radiolabeled Molecules

2017

This work describes methods to establish acute and chronic hyperglycemia models in zebrafish. The aim is to investigate the impact of hyperglycemia on physiological processes, such as constitutive and injury-induced neurogenesis. The work also highlights the use of zebrafish to follow radiolabeled molecules (here, [18F]-FDG) using PET/CT.

Hyperglycemia is an abnormally high blood glucose level. It is diagnosed by fasting glucose ≥126 mg/dL, 2-hour oral glucose tolerance test (or OGTT) ≥200 mg/dL, random glucose ≥200 mg/dL with symptoms, or HbA1c ≥6.5%. However, HbA1c results may be unreliable in certain conditions, such as anemia or hemoglobinopathies, and the diagnosis should be confirmed unless classic symptoms are present. Postprandial hyperglycemia is typically considered significant when glucose levels exceed 180 mg/dL two...

Renal injuries incurred from nephrotoxins, which include drugs ranging from antibiotics to chemotherapeutics, can result in complex disorders whose pathogenesis remains incompletely understood. This protocol demonstrates how zebrafish can be used for disease modeling of these conditions, which can be applied to the identification of renoprotective measures.
