Cell-free Ecm Sheets

Cell-free ECM sheets are tissue-derived extracellular matrix scaffolds stripped of cellular components while preserving the three-dimensional architecture and biochemical cues that support tissue organization. During decellularization, cells and nucleic acids are removed while structural proteins, matrix-bound factors, and native microstructure remain available for interaction with newly introduced cells. In developmental biology, these sheets provide a biomimetic platform for studying cell adhesion, migration, proliferation, and differentiation within a tissue-like environment. They also support investigations of extracellular signals that regulate morphogenesis and may inform engineered tissues, regenerative strategies, and models of developmental disorders.

A method to obtain nanofibers and complex nanostructures from single or multiple extracellular matrix proteins is described. This method uses protein-surface interactions to create free-standing protein-based materials with tunable composition and architecture for use in a variety of tissue engineering and biotechnology applications.

This combinatorial design incorporates organ-specific layered cell sheets with two distinct biologically-derived materials containing a strong fibrous matrix base, and endothelial cells for enhancing new vessels formation.
