Lymph Gland Mounting

Lymph gland mounting is a specimen-preparation method that preserves and positions lymph glands for microscopic analysis, making developmental changes in hematopoietic tissues visible. In this procedure, dissected glands are typically fixed, stained, and oriented in mounting medium beneath a coverslip so that their cellular architecture and molecular markers can be imaged. In developmental biology, mounted lymph glands enable researchers to examine progenitor populations, differentiated blood-cell zones, cell division, and signaling pathways during organ development. The resulting preparations support fluorescence microscopy and quantitative analysis of hematopoiesis, helping clarify how blood-cell formation is organized and regulated.

Methods to Examine the Lymph Gland and Hemocytes in Drosophila Larvae

Drosophila and mammalian hematopoietic systems share many common features, making Drosophila an attractive genetic model to study hematopoiesis. Here we demonstrate dissection and mounting of the major larval hematopoietic organ for immunohistochemistry. We also describe methods to assay various larval hematopoietic compartments including circulating hemocytes and sessile crystal cells.

We developed a method to successfully remove, process, section, and stain, for histopathological evaluation, mammary tissue that had originally been fixed on slides as whole mounts. This method may promote the collection and evaluation of mammary gland whole mounts in reproductive and developmental test guideline studies.

Mammary gland development in the rodent has typically been evaluated using descriptive assessments or by measuring basic physical attributes. Branching density is an indicator of mammary development that is difficult to quantify objectively. This protocol describes a reliable method for the quantitative assessment of mammary gland branching characteristics.

High-frequency Ultrasound Imaging of Mouse Cervical Lymph Nodes

This protocol describes the application of high-frequency ultrasound (HFUS) for imaging mouse cervical lymph nodes. This technique optimizes visualization and quantification of cervical lymph node morphology, volume and blood flow. Image-guided biopsy of cervical lymph nodes and processing of lymph tissue for histological evaluation is also demonstrated.
