Somitogenesis Heart Formation

Somitogenesis is the embryonic process that segments paraxial mesoderm into regularly spaced somites, establishing the body’s axial organization and guiding coordinated tissue development. A molecular segmentation clock, involving rhythmic gene expression and a wavefront of FGF, Wnt, and retinoic acid signaling, determines when cells separate into epithelial somites that later form vertebrae, ribs, and skeletal muscle. Although the heart arises primarily from lateral plate mesoderm, its formation is coordinated with somitogenesis through shared positional cues and tissue interactions that influence cardiac progenitor organization, heart tube formation, and morphogenesis. Studying this relationship clarifies congenital malformations and supports developmental and regenerative biology research.

Developmental studies in the mouse are hampered by the inaccessibility of the embryo during gestation. To promote the long-term culture of the embryonic heart at late stages of gestation, we developed a protocol in which the excised heart is cultured in a semi-solid, dilute Matrigel.

The cuff technique shortens and facilitates the model of heterotopic cervical heart transplantation in mice by avoiding technically challenging suture anastomoses of small vessels. The technical details shown in this video paper should allow researches to establish this model in their laboratories.

We have developed a Semi-automated Optical Heartbeat Analysis method (SOHA) for analyzing high speed optical recordings from Drosophila, zebrafish and embryonic mouse hearts. We demonstrate the application of our methodology to the analysis of heart function in fruit fly and embryonic mouse hearts.
