Meckel's Cartilage

Meckel’s cartilage is an embryonic cartilage rod that forms within the first pharyngeal arch and guides development of the vertebrate jaw and related craniofacial structures. During mandibular development, neural crest-derived cells organize and chondrify along this rod, while surrounding mesenchyme forms much of the mandible through intramembranous ossification rather than by replacing the cartilage directly. Most of Meckel’s cartilage later regresses, but its dorsal region contributes to the malleus and incus, and associated tissues form structures such as the sphenomandibular ligament. Studying this transient scaffold clarifies cartilage remodeling, jaw morphogenesis, and the developmental origins of craniofacial abnormalities.

Cell organization of craniofacial bones has long been hypothesized but never directly visualized. Multi-spectral cell labeling and in vivo live imaging allows visualization of dynamic cell behavior in zebrafish lower jaw. Here, we detail the protocol to manipulate Zebrabow transgenic fish and directly observe cell intercalation and morphological changes of chondrocytes in the Meckel’s cartilage.

The methods described in this paper show how to convert a commercial inkjet printer into a bioprinter with simultaneous UV polymerization. The printer is capable of constructing 3D tissue structure with cells and biomaterials. The study demonstrated here constructed a 3D neocartilage.

Goal of the presentation is to demonstrate a highly reproducible method to generate matrix associated stem cell implants in cartilage defects, which can be visualized with MR imaging. Stem cells are labeled with FDA-approved Ferumoxides, mixed with agarose, implanted into cartilage defects and imaged with a 7T MR scanner.
