Mesenchymal Stem Cell Isolation

Mesenchymal stem cell isolation is the laboratory process of separating mesenchymal stem cells from tissues such as bone marrow, adipose tissue, or other stromal sources for study and use in regenerative research. Typically, tissue is mechanically disrupted and enzymatically digested to release cells, which are then enriched through differential adherence to plastic or separation methods based on surface markers and physical properties. In developmental biology, isolated mesenchymal stem cells provide a model for examining lineage commitment, tissue formation, and interactions within developing microenvironments. Reliable isolation and characterization are essential for studying osteogenic, chondrogenic, and adipogenic differentiation and for comparing developmental potential across tissue sources.

Rat MSCs were isolated from femurs and tibias and then enriched by magnetic cell sorting. Sorted cells were confirmed for the expression of surface markers by flow cytometry. These cells were also cultured at clonal density to form single colonies and then these colonies were separated by cloning cylinders.

Here, we describe a method for the isolation of cells in the pancreas microenvironment from embryonic, neonatal and adult mouse tissue, focusing on the isolation of mesenchymal cells. This method allows profiling of cell gene expression and protein secretion in order to elucidate the extrinsic signals that regulate pancreas development, function, and tumorigenesis.
