Murine Lung Mscs

Murine lung MSCs are mouse-derived mesenchymal stromal cells found in the lung’s connective-tissue compartment, where they contribute to tissue organization, repair, and homeostasis during development. In culture, these cells are typically enriched from dissociated lung tissue by adherence and expansion, then identified through characteristic marker profiles and their capacity for mesenchymal differentiation; in vivo, they influence neighboring epithelial and vascular cells through extracellular-matrix production and paracrine signaling. Developmental biologists use murine lung MSCs to investigate mesenchyme–epithelium interactions, alveolar formation, injury responses, and fibrotic remodeling. These studies help clarify how stromal populations shape organ development and may inform regenerative research.

Isolation and Enrichment of Rat Mesenchymal Stem Cells (MSCs) and Separation of Single-colony Derived MSCs

Rat MSCs were isolated from femurs and tibias and then enriched by magnetic cell sorting. Sorted cells were confirmed for the expression of surface markers by flow cytometry. These cells were also cultured at clonal density to form single colonies and then these colonies were separated by cloning cylinders.

The immunomodulatory properties of human mesenchymal stem cells (MSC) appear increasingly relevant for clinical application. Using a co-culture system of MSCs and peripheral blood leukocytes pre-stained with the fluorescent dye carboxyfluorescein succinimidyl ester (CFSE), we describe the in vitro assessment of MSC immunomodulation on effector leukocyte proliferation and specific subpopulations.

This video demonstrates an in vitro technique to identify immune cells residing in murine lungs. After isolating target immune cells from murine lung tissue, they are surface-labeled using a cocktail of fluorophore-tagged antibodies against specific markers on innate and adaptive immune cells. Additionally, monocytes, macrophages, and dendritic cells are labeled using an intracellular marker. The various immune cell types are then identified using flow cytometry.
