Primary Retinal Culture

Primary retinal culture is an ex vivo method for maintaining cells or tissue taken directly from the retina under controlled laboratory conditions, providing a model for studying retinal development. Researchers isolate retinal tissue, dissociate or maintain it as an explant, and support cell survival and differentiation in a nutrient-defined culture environment that can preserve aspects of native cell signaling and organization. In developmental biology, these cultures help investigate neurogenesis, photoreceptor and retinal neuron differentiation, axon growth, and interactions among retinal cell types. They also enable controlled manipulation of growth factors, genes, and environmental conditions to clarify mechanisms that shape retinal formation and function.

Generation of Primary Retinal Ganglion Cell Cultures From Zebrafish Embryos

2025

The video demonstrates a method for culturing primary retinal ganglion cells from zebrafish embryos. Early-stage embryos undergo dissection to isolate the eyes, which are subsequently mechanically dissociated into a suspension of retinal ganglion cells. This cell suspension is cultured on coverslips coated with adhesive proteins to facilitate axon development.

This video article demonstrates the establishment of organotypic retinal wholemount cultures and a cytospin procedure for analysis of exogenously induced effects. Organotypic retinal wholemount cultures mimic the in vivo situation and significantly facilitate the accessibility of murine retinas for experimental manipulations while circumventing the disadvantages of classical murine animal models.
