Rpe Marker Expression

RPE marker expression refers to the presence and level of molecular markers that identify retinal pigment epithelium (RPE) cells, providing evidence of their identity and developmental state. During eye development, regulatory factors such as OTX2 and MITF activate RPE-associated genes, including RPE65, BEST1, PMEL, and genes involved in pigmentation, maturation, and epithelial function. Assessing these markers with methods such as immunostaining, gene-expression analysis, or microscopy helps distinguish RPE cells from related retinal populations and track differentiation in stem-cell and organoid models. This analysis supports studies of retinal development, disease modeling, cell-based therapies, and the quality control of engineered RPE tissues.

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.
