Metabolite Profiling Methods

Metabolite profiling methods are analytical approaches used to measure and compare the small molecules produced or transformed by cells, organisms, and environmental systems. These methods typically extract metabolites from a sample, separate them by chromatography or related techniques, and identify or quantify them using mass spectrometry or nuclear magnetic resonance spectroscopy. In environmental research, metabolite profiles can reveal how organisms respond to pollutants, temperature shifts, nutrient availability, or other ecological pressures. Comparing profiles across samples helps identify biochemical pathways, assess ecosystem health, track contaminant effects, and generate biomarkers for environmental monitoring.

Detection of Human Fecal Metabolites Using a Bacterial Biosensor

Begin with a bacterial biosensor culture suspended in a medium. These engineered bacteria produce a transcription factor that, upon binding to a specific fecal metabolite, activates a promoter associated with a fluorescent reporter gene.To this culture, add a filtered human fecal sample containing metabolites, such as lactate, that are involved in host-microbiome interactions.Dilute this mixture with a fresh medium to maintain an optimal reaction volume.Incubate without shaking to mimic the...
