Formaldehyde Triton X-100

Formaldehyde Triton X-100 treatment combines chemical fixation with membrane permeabilization to prepare cells or tissues for genetic and cytological analysis. Formaldehyde creates covalent cross-links that preserve cellular structures and nucleic acid organization, while Triton X-100 disrupts lipid membranes so probes, antibodies, or other detection reagents can enter the cell. This combination supports fluorescence in situ hybridization, immunostaining, and related microscopy-based methods used to examine chromosomes, genomic loci, and nuclear organization. By maintaining spatial relationships while improving reagent access, it helps researchers connect genetic information with its location and behavior within cells.

This video demonstrates the immunohistochemical staining for S-100 protein in rectal suction biopsy sections. Antigen-retrieved sections are blocked and treated with primary, followed by secondary antibodies against S-100, a marker of Hirschsprung's disease. Chromogenic substrate is added to stain the targeted cells. Samples are dehydrated and mounted before visualizing under the microscope.
