Host Pathway Identification

Host Pathway Identification is the process of determining which cellular genes and molecular pathways influence a host’s response to infection, disease, or other biological conditions. In genetics, researchers perturb host genes using approaches such as CRISPR screens or RNA interference, measure changes in a defined phenotype, and use pathway-enrichment analysis to connect individual genes with processes such as signal transduction, membrane trafficking, or innate immune defense. This approach reveals host factors required for pathogen entry, replication, or clearance and helps distinguish protective responses from pathways that promote disease. Its findings can guide studies of gene function, biomarker development, and host-directed therapeutic strategies.

Here, we present the protocols to identify 1) virus-encoded immunomodulators that promote arbovirus replication and 2) eukaryotic host factors that restrict arbovirus replication. These fluorescence- and luminescence-based methods allow researchers to rapidly obtain quantitative readouts of arbovirus replication in simplistic assays with low signal-to-noise ratios.
