Mir-125b Parp-1 Interaction

The miR-125b–PARP-1 interaction describes regulatory crosstalk between miR-125b, a small noncoding RNA, and poly(ADP-ribose) polymerase 1 (PARP-1), a protein involved in DNA damage responses and genome maintenance. miR-125b can recognize complementary sequences in PARP-1 messenger RNA, typically within its 3′ untranslated region, leading to transcript degradation or reduced protein translation; changes in miR-125b levels can therefore alter PARP-1 activity. Studying this interaction helps clarify post-transcriptional gene regulation, DNA repair, apoptosis, and cellular responses to genomic stress, with potential relevance to cancer biology and the development of biomarkers or targeted therapies.

Here, we describe a method for cell-based visualization, subcellular localization, and quantitative analysis of poly(ADP-ribose) (PAR) enriched foci in fixed mammalian cells. This assay enables the quantification of sites of DNA damage-induced PARP activation, including that associated with base excision repair or single-strand break repair, in the nuclei of genotoxin-exposed cells.

This manuscript describes the efficient, non-viral delivery of miR to endothelial cells by a PEI/MNP vector and their magnetization. Thus, in addition to genetic modification, this approach allows for magnetic cell guidance and MRI detectability. The technique can be used to improve the characteristics of therapeutic cell products.
