Nematode Screening

Nematode screening is a genetic method that uses nematodes, especially Caenorhabditis elegans, to identify mutations or gene activities that produce a measurable phenotype. Researchers generate or collect genetically varied worms, expose them to defined conditions, and examine traits such as development, movement, fertility, morphology, or survival, often using microscopy and controlled crosses to link phenotypes with genetic changes. Because nematodes reproduce rapidly and support large, comparatively simple populations, screening can reveal genes and pathways involved in development, behavior, stress responses, and disease-related biology. The approach supports forward and reverse genetics, functional gene analysis, and efficient testing of candidate mutations.

Screening Cotton Genotypes for Reniform Nematode Resistance

Here, a protocol is presented for the rapid non-destructive screening of cotton genotypes for reniform nematode resistance. The protocol involves visually examining the roots of nematode-infected cotton seedlings to determine infection response. The vegetative shoot from each plant is then propagated to recover plants for seed production.

Visualizing Bacteria in Nematodes using Fluorescent Microscopy

To study the mutualism between Xenorhabdus bacteria and Steinernema nematodes, methods were developed to monitor bacterial presence and location within nematodes. The experimental approach, which can be applied to other systems, entails engineering bacteria to express the green fluorescent protein and visualizing, using fluorescence microscopy bacteria within the transparent nematode.

Here we present several protocols for quantifying predatory feeding behavior found in the model nematode Pristionchus pacificus and its relatives. Additionally, we provide methods for analyzing predatory feeding adaptations including mouth structures and teeth.
