Cardiac Ecm Isolation

Cardiac ECM isolation is the process of separating the extracellular matrix from heart tissue to study its structural, biochemical, and biological properties. It commonly uses controlled tissue processing, including removal of cellular material while preserving matrix components such as collagen, elastin, glycoproteins, and proteoglycans. In immunology and infection research, isolated cardiac ECM provides a defined model for examining how matrix signals influence immune-cell adhesion, migration, and activation, as well as how tissue structure changes during inflammation or pathogen-associated injury. These preparations also support biomaterial development and investigations of cardiac repair.

Isolation of Functional Cardiac Immune Cells

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.

Adult cardiac myocytes are primary cells that can be isolated from animal hearts and cultured for several days. Within this culture period adenoviral gene transfer can be used to express genetically encoded biosensors (GEBs) or fluorescent fusion proteins. Both approaches allow cellular investigations by means of confocal microscopy.

Langendorff-mode isolated heart perfusion, in conjunction with 31P NMR spectroscopy, combines the fields of biochemistry and physiology into one experiment. The protocol allows for the dynamic measurement of high energy phosphate content and turnover in the heart while concurrently monitoring physiologic function. When performed correctly, this is a valuable technique in the assessment of cardiac energetics.

A method to obtain nanofibers and complex nanostructures from single or multiple extracellular matrix proteins is described. This method uses protein-surface interactions to create free-standing protein-based materials with tunable composition and architecture for use in a variety of tissue engineering and biotechnology applications.
