Cd11b

CD11b is the alpha M subunit of integrin Mac-1, a leukocyte surface receptor that helps immune cells recognize, adhere to, and respond to inflammatory cues. It pairs with CD18 to form complement receptor 3 (CR3), which binds complement-opsonized particles and promotes firm adhesion, migration, and phagocytosis through cytoskeletal signaling. Expressed on neutrophils, monocytes, macrophages, and other myeloid cells, CD11b links innate immune surveillance with pathogen clearance and regulation of inflammation. Measuring or manipulating CD11b can therefore help characterize leukocyte activation, study host-pathogen interactions, and investigate inflammatory and immunodeficiency disorders.

2017

Here, we present a protocol to isolate microglia from postnatal mouse pups (day 1) for in vitro experimentation. This improvised method of isolation generates both high yield and purity, a significant advantage over alternate methods that allows broad range experimentation for the purposes of elucidating microglial biology.

Magnetic Isolation of Microglia from Mouse Pup Brains

2025

In this video, we describe a protocol for isolating CD11b cells, including microglia, from mouse pup brains using magnetic-activated cell sorting. The brain tissues are homogenized to obtain single-cell suspensions, which are incubated with anti-CD11b antibody-coated microbeads, followed by a magnetic column-based separation to purify the CD11b+ cell population.

Measuring Microglial Uptake of Intravitreally-Delivered Fluorescent Particles Using Flow Cytometry

2025

This video demonstrates flow cytometry for measuring microglial uptake of fluorescent bioparticles after intravitreal delivery in a mouse eye. Flow cytometry detects live cells expressing CD11b and distinguishes phagocytic microglia cells exhibiting green fluorescence from other myeloid cells, including Ly6C-expressing monocytes and Ly6G-expressing neutrophils.

Isolating Microglia from Mouse Brain Demyelinating Lesions Using Magnetic Activated Cell Sorting

2025

This video demonstrates a protocol for extracting microglial cells from demyelinating lesions in mouse brains using magnetic cell sorting. The protocol involves dissecting the lesioned brain tissue containing microglial cells, treating the tissue with specific enzymes to obtain a single-cell suspension, and utilizing microglial receptor-specific magnetic beads to isolate the microglial cells.

In Vitro Generation of Plasmacytoid Dendritic Cells from Common Lymphoid Progenitors

2025

This video demonstrates an in vitro assay to generate plasmacytoid dendritic cells (pDCs) from common lymphoid progenitor cells (CLPs). Upon co-culturing purified CLPs with γ-irradiated feeder cells in the presence of Fms-like tyrosine kinase-3 ligand (FL), the CLPs differentiate into non-adherent pDCs, the presence of which is confirmed using flow cytometry.
