Cfse Staining

CFSE staining is a fluorescent cell-labeling technique used to track cell division, making it valuable for measuring immune-cell proliferation during infection and immune responses. The membrane-permeable dye enters cells and is converted into a reactive form that binds intracellular amines; as labeled cells divide, the fluorescence is distributed between daughter cells and decreases in distinct steps. Flow cytometry can detect these fluorescence peaks to quantify proliferation, identify responding cell populations, and estimate division history. In immunology and infection research, CFSE staining helps assess lymphocyte activation, antigen-specific responses, and the effects of pathogens, vaccines, or immunomodulatory treatments.

Many infections elicit a strong CTL response, but occasionally, the quantity of responding cells does not correlate to control of the pathogen1. One measure of CTL quality is their ability to kill specifically2. CFSE labeling of target cells can be used to investigate this CTL response quality in vivo3,4.

CFSE covalently labels long-lived intracellular molecules with the fluorescent dye, carboxyfluorescein. As such, when a CFSE-labeled cell divides, its progeny have half the amount of fluorescence, which can thereby be used to assess cell division. This article describes the procedures typically used for labeling mouse lymphocytes with CFSE.
