Dual Color Elispot

Dual Color ELISpot is an immunoassay that measures the secretion of two analytes from individual cells, providing a functional view of immune responses rather than relying only on cell counts. In this technique, antigen-stimulated or otherwise activated cells release cytokines onto a capture-antibody-coated membrane; two detection systems then bind their targets and generate distinct colored spots, whose number reflects responding cell frequency and whose color pattern indicates single or dual secretion. In immunology and infection research, Dual Color ELISpot helps compare pathogen-specific T-cell responses, characterize multifunctionality, evaluate vaccines, and monitor immune responses during infection or treatment.

Functional Assessment of Antibody-Secreting B Cells Using ELISpot Assay

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.
