Etheno Adduct Analysis

Etheno adduct analysis is a set of analytical approaches used to detect and quantify etheno-modified nucleobases, DNA lesions formed when reactive aldehydes generated during lipid peroxidation or inflammation react with DNA. The process typically involves isolating DNA or biological samples, enzymatically releasing modified nucleosides, and measuring them with immunochemical methods or liquid chromatography coupled to mass spectrometry, often against authentic standards. In immunology and infection research, these adducts provide molecular evidence of oxidative and electrophilic stress, helping link inflammatory responses or pathogen-associated damage to genotoxic effects. Measurements can support biomarker development, exposure assessment, and evaluation of antioxidant or anti-inflammatory interventions.

This article describes a novel protocol and reagent set designed for sensitive measurement of neurotoxic effects of compounds and treatments on co-cultures of neurons and astrocytes using high content analysis.
