Toxin Crystal Purification

Toxin crystal purification is a laboratory process that isolates toxin molecules in crystalline form, reducing accompanying proteins, nucleic acids, and other soluble impurities. Toxin molecules are concentrated until supersaturation promotes nucleation and crystal growth; the resulting crystals can be collected and washed to remove contaminants while preserving the target toxin. In immunology and infection research, purified toxin crystals provide consistent material for structural analysis, receptor or antibody binding studies, activity testing, and investigations of host-pathogen interactions. These preparations can improve experimental reproducibility and help clarify how toxins cause disease or stimulate protective immune responses.

This manuscript describes how to screen for thermostabilizing mutations, purify the human serotonin transporter, generate high affinity antibodies, and crystallize the serotonin transporter-antibody complex bound to the antidepressant drug S-citalopram. This protocol can be adapted to the study of other challenging membrane transporters, receptors, and channels.
