Viral Rna Quantification

Viral RNA quantification is the measurement of viral RNA in a sample, providing a molecular estimate of viral burden that supports diagnosis, infection monitoring, and research in immunology and infection. In a common approach, reverse transcription converts RNA into complementary DNA, which quantitative PCR amplifies while fluorescence records cycle-dependent signal; comparison with standards can translate that signal into an RNA concentration or copy number. The resulting measurements help characterize replication dynamics, evaluate host immune responses, and assess changes during treatment or experimental infection. Careful sample preparation and controls are essential for distinguishing true viral signal from contamination or technical variation.

Detection of Viral RNA by Fluorescence in situ Hybridization (FISH)

A fluorescence in situ hybridization (FISH) method was developed to visually detect viral genomic RNA using fluorescence microscopy. A probe is made with specificity to the viral RNA that can then be identified using a combination of hybridization and immunofluorescence techniques. This technique offers the advantage of identifying the localization of the viral RNA or DNA at steady-state, providing information on the control of intracellular virus trafficking events.
