Cd3/cd8 Gating Strategy

The CD3/CD8 gating strategy is a flow cytometry method for identifying and quantifying CD8-positive T lymphocytes within a mixed cell sample, an important measurement in immunology and infection research. After excluding debris, doublets, and nonviable cells, analysts gate CD3-positive events to define the T-cell population, then assess CD8 expression to resolve the CD3+CD8+ subset. This sequential approach helps distinguish cytotoxic T cells from other leukocytes and supports accurate comparison of immune-cell frequencies across samples. It is used to evaluate cellular immune responses, monitor changes during infection, and characterize treatment- or vaccine-associated immunological outcomes.

This article describes the methods for screening the genes controlling plasmodesmal permeability and hence auxin gradient during tropic response. This includes the measurement of the degree of tropic response in hypocotyl of Arabidopsis thaliana and checking plasmodesmal permeability by 8-hydroxypyrene-1,3,6-trisulfonic acid (HPTS) loading and finally callose level assessment.

Ligand-Gated Ion Channel Receptor: Gating Mechanism

2023

Ligand-gated ion channels are transmembrane proteins that play a vital role in intercellular communication and functions of the nervous system. They allow the influx of ions across the membrane once the neurotransmitter binds, allowing the subsequent transmission of electrical excitation across the neurons. Other ligand-gated ion channels, like the γ-aminobutyric acid (GABA) receptor, permit anions like chloride into the cells on the binding of the GABA molecule. Their entry into the cell...

Underflow Gates

2025

Underflow gates are vital for controlling water flow in irrigation canals. The three main types of underflow gates — vertical, radial, and drum gates — serve different purposes while ensuring effective flow management. Vertical gates move up and down, generating a free-flowing water jet; radial gates pivot to regulate the flow; and drum gates rotate for precise adjustments. The flow through these gates is influenced by downstream conditions, resulting in free or drowned outflow.Free and Drowned...
