Blood Volume Measurement

Blood volume measurement is the quantitative assessment of the circulating blood within the vascular system, including plasma and cellular components, and is important for evaluating fluid balance and circulatory status. In indicator-dilution methods, a known amount of a labeled tracer is introduced into the bloodstream, allowed to mix, and measured in a blood sample; the tracer concentration then provides the distribution volume, often with separate tracers for plasma and red blood cells. In medicine, these measurements help identify hypovolemia, polycythemia, anemia, and abnormal fluid shifts when routine vital signs or hematocrit alone are insufficient. The results can guide diagnosis, treatment planning, and assessment of physiological changes.

Here we present a protocol to simply and reliably measure the lung pressure-volume curve in mice, showing that it is sufficiently sensitive to detect phenotypic parenchymal changes in two common lung pathologies, pulmonary fibrosis and emphysema. This metric provides a means to quantify the lung’s structural changes with developing pathology.
