Brain Tumor Surgery

Brain tumor surgery is a neurosurgical procedure used to diagnose, remove, or reduce tumors in the brain while preserving healthy tissue and neurological function. Surgeons may perform a biopsy or craniotomy, using magnetic resonance imaging, neuronavigation, and, when appropriate, awake brain mapping to guide access and identify critical functional areas. Resection can relieve pressure, reduce symptoms, and provide tissue for pathological classification, which informs radiation, chemotherapy, or targeted treatment. Because tumor location, type, and relationship to vital structures vary widely, surgical planning requires a careful balance between removing abnormal tissue and limiting neurological injury.

In early gastric cancer, the aim of surgery is to precisely remove the distal stomach including the primary tumor. To do this, accurate localization of the tumor is crucial, especially in totally laparoscopic surgery. This protocol describes a procedure for intraoperative gastroscopy in totally laparoscopic subtotal gastrectomy.

Targeted ablation of specific brain region(s) by infusion of an excitotoxin using stereotaxic coordinates is described. This technique could also be adapted for infusion of other chemicals into the rat brain.

Luciferase-modified human brain tumor xenografts can be established intracranially in athymic mice, with subsequent monitoring of tumor growth and response to therapy using bioluminescence imaging. In combination with survival analysis, bioluminescence monitoring is an essential research tool for pre-clinical testing of therapies being considered for treating brain tumors.

This video and protocol demonstrate how to implant a glass-covered cranial window in rodents. These preparations can be used for chronic in vivo two-photon imaging of the neocortex over time scales of months. It may also be used for other types of imaging, including optical intrinsic signal imaging.
