Cardiac Inflammation

Cardiac inflammation is an immune response affecting the heart or its surrounding tissues, and it can disrupt normal cardiac function. It may arise after infection, tissue injury, or autoimmune activity, when inflammatory mediators recruit immune cells and cause swelling that can impair myocardial contraction, electrical conduction, or pericardial movement. In medicine, recognizing conditions such as myocarditis and pericarditis supports evaluation with clinical assessment, electrocardiography, cardiac imaging, and laboratory biomarkers. Understanding the underlying cause guides treatment and helps clinicians monitor risks including arrhythmias, reduced cardiac output, heart failure, and recurrent inflammation.

Streptococcus pneumoniae forms discrete non-purulent microscopic lesions in the heart. Outlined is the protocol for a murine model of cardiac microlesion formation. Instruction is provided on microlesion visualization using microscopy, discrimination between early and late microlesions, and methods to detect cardiac remodeling in hearts of convalescent animals.

We describe the protocol to perform a cardiac stress test induced by dobutamine and monitored by cardiac catheterization in normal mice. Also we show its application to unmask subclinical cardiac disease in high fat diet-induced obese mice.

In vivo Imaging Method to Distinguish Acute and Chronic Inflammation

We describe a non-invasive imaging method for distinguishing inflammatory stages. Systemic delivery of luminol reveals areas of acute inflammation dependent upon MPO activity in neutrophils. In contrast, injection of lucigenin allows for visualization of chronic inflammation dependent upon Phox activity in macrophages.

Creation of Cardiac Tissue Exhibiting Mechanical Integration of Spheroids Using 3D Bioprinting

This protocol describes 3D bioprinting of cardiac tissue without the use of biomaterials. 3D bioprinted cardiac patches exhibit mechanical integration of component spheroids and are highly promising in cardiac tissue regeneration and as 3D models of heart disease.
