Cortical Penetration

Cortical penetration is the extent to which a therapeutic substance reaches the cerebral cortex, a key consideration in treating disorders that affect brain tissue. It depends on factors such as the blood-brain barrier, molecular size and properties, blood flow, tissue diffusion, and the delivery route, which together determine how effectively a compound enters and spreads through cortical regions. Understanding these processes supports the development and evaluation of medicines for conditions including brain tumors, epilepsy, stroke, and neuroinflammatory disease. Measuring cortical penetration can also guide drug formulation, dosing strategies, and targeted delivery approaches in neurological research.

This protocol describes the synthesis of cyclic cell-penetrating peptides with aromatic cross-links and the evaluation of their permeability across biological barriers.

Shifting Zebrafish Lethal Skeletal Mutant Penetrance by Progeny Testing

The goal of this protocol is to alter the penetrance of lethal skeletal mutant phenotypes in zebrafish by selective breeding. Lethal mutants cannot be grown to adulthood and bred themselves, therefore this protocol describes a method for tracking and selecting penetrance through multiple generations by progeny testing.

Inducing Penetrating Traumatic Brain Injury in Adult Drosophila Flies

2025

This video demonstrates the induction of penetrating traumatic brain injury in the drosophila brain. Newly emerged flies are selected, anesthetized, and positioned under a fluorescence-equipped stereo microscope. The GFP-expressing mushroom body is identified, and a Minutien pin is inserted to cause injury. The flies are allowed to recover for further studies.

Subtype-selective Electroporation of Cortical Interneurons

2014

This procedure shows how to target interneurons in the developing mouse forebrain by means of in utero electroporation. This technique was particularly efficient to achieve selective gene expression in interneuron subtypes destined to the superficial layers of the cortex.

Fluorescence Leakage Assay to Study Cell-Penetrating Peptide Interaction with Membrane

2025

This video discusses the fluorescence leakage assay to study the interactions of cell-penetrating peptides with cell membranes using cell membrane mimics — large unilamellar vesicles, or LUVs, encapsulated with a fluorescent dye and a quencher.
