Drug Delivery Hydrogel

Drug delivery hydrogels are water-rich, three-dimensional polymer networks designed to carry and release therapeutic agents at a controlled rate, making them useful for localized and sustained treatment. Their crosslinked structure absorbs biological fluids, while drug movement is governed by diffusion, hydrogel swelling, polymer degradation, or combinations of these processes. Formulations can be engineered to respond to conditions such as pH, temperature, enzymes, or other signals, helping regulate when and where a drug is released. In medicine, these materials support localized therapy, reduce dosing frequency, and may improve treatment effectiveness while limiting systemic exposure in applications such as wound care, cancer therapy, and tissue repair.

Supramolecular hydrogelators based on ureido-pyrimidinones allow full control over the macroscopic gel properties and the sol–gel switching behavior using pH. Here, we present a protocol for formulating and injecting such a supramolecular hydrogelator via a catheter delivery system for local delivery directly in relevant areas in the pig heart.
