Facial Motor Nucleus

The facial motor nucleus is a cluster of lower motor neurons in the pons that controls the muscles of facial expression, making it essential for communication, swallowing, eye protection, and other functions. Its neurons receive corticobulbar input from the cerebral cortex and send axons through the facial nerve to activate muscles on the same side of the face; cortical control is typically bilateral for the upper face and predominantly contralateral for the lower face. In medicine, this organization helps clinicians localize lesions and distinguish central causes, such as stroke, from peripheral facial nerve disorders, including Bell’s palsy, supporting diagnosis, prognosis, and treatment planning.

We present a surgical protocol detailing how to perform a cut or crush axotomy on the facial nerve in the mouse. The facial nerve axotomy can be employed to study the physiological response to nerve injury and test therapeutic techniques.

A protocol for separation of embryo facial ectoderm and mesenchyme is described. We use Dispase II to treat whole embryos first, dissect whole facial prominences out, and then separate the facial ectoderm and mesenchyme.

Mouse Hindbrain Ex Vivo Culture to Study Facial Branchiomotor Neuron Migration

Embryonic neurons are born in the ventricular zone of the neural tube, but migrate to reach appropriate targets. Facial branchiomotor (FBM) neurons are a useful model to study neuronal migration. This protocol describes the wholemount ex vivo culture of mouse embryo hindbrains to investigate mechanisms that regulate FBM migration.
