Human Tumor Cells

Human tumor cells are abnormal human cells that acquire genetic and functional changes allowing them to grow, survive, and sometimes spread beyond normal tissue boundaries. These changes can disrupt cell-cycle regulation, growth signaling, DNA repair, and programmed cell death, enabling uncontrolled proliferation and, in malignant tumors, invasion or metastasis. In medicine, researchers study tumor cells in patient samples, cultured cell lines, organoids, and animal models to investigate cancer biology and evaluate diagnostic methods, drug responses, and treatment resistance. This work supports the development of targeted therapies and improves understanding of how tumors evolve within their tissue environment.

We present a simple agarose overlay platform to grow 3D multicellular spheroids using neuroendocrine cancer cell lines. This method provides a very convenient way to examine the effect of therapeutic drugs on the neuroendocrine tumor cells. It could also help us establish human neuroendocrine tumor spheroids for cancer therapy.

Human tumor xenografts are vascularized and infiltrated by cells of mouse origin during the growth phase in vivo. To circumvent the bias caused by these contaminating cells during downstream analysis, we developed a method allowing for comprehensive depletion of all mouse cells by magnetic cell sorting.

The generation and characterization of tumor specific T cells using humanized mice is described here. Human thymic tissue and genetically modified human hematopoietic stem cells are transplanted into immunocompromised mice. This results in the reconstitution of an engineered human immune system allowing for in vivo examination of anti-tumor immune responses.
