Non-anticoagulated Blood

Non-anticoagulated blood is blood collected without chemical agents that inhibit coagulation, allowing its natural clotting response to occur. After collection, clotting factors activate thrombin, which converts fibrinogen to fibrin and forms a clot that separates cellular components from serum. In medicine, this material supports serum-based laboratory testing, blood-clot evaluation, and studies of hemostasis, the process that limits bleeding after vascular injury. Its use can provide clinically relevant information about coagulation and tissue injury, although handling conditions and processing time must be controlled because clot formation changes the composition of the sample.

We present an in vitro model which allows the study and analysis of coagulation in whole, non-anticoagulated blood. Anticoagulation in the system depends on the natural anticoagulation effect of healthy endothelial cells and endothelial cell activation will result in clotting.

Rapid Point-of-Care Assay of Enoxaparin Anticoagulant Efficacy in Whole Blood

Demonstration of a rapid quantitative assay of the inhibition of blood coagulation by the low-molecular-weight-heparin, enoxaparin. The contribution of enoxaparin is assessed by removing its influence through digestion with heparinase. A fuller description of the assay is detailed in our published paper.1 The assay still requires clinical confirmation.

Anticoagulant Drugs: Vitamin K Antagonists and Direct Oral Anticoagulants

2024

Oral anticoagulants are vital tools in preventing and treating blood clotting disorders. This diverse class of medications can be categorized as vitamin K antagonists, exemplified by warfarin, and direct thrombin inhibitors (DTIs), such as dabigatran, as well as factor Xa inhibitors, including rivaroxaban. Warfarin, a prominent vitamin K antagonist family member, exerts its effect by inhibiting the enzyme VKORC1 (vitamin K epoxide reductase complex 1). By hindering this enzyme, warfarin...

Source: Kay Stewart, RVT, RLATG, CMAR; Valerie A. Schroeder, RVT, RLATG. University of Notre Dame, IN Blood collection is a common requirement for research studies that involve mice and rats. The method of blood withdrawal in mice and rats is dependent upon the volume of blood needed, the frequency of the sampling, the health status of the animal to be bled, and the skill level of the technician. All methods discussed-retro-orbital sinus bleeds, initial tail snip bleeds, and intracardiac...

Here, we describe a simple tail vein blood sampling method in non-anesthetized rats using a vacuum extraction tube system. This method reduces the risk of direct exposure to blood and simplifies taking multiple samples from a single venipuncture.
