Pathway Activity Monitoring

Pathway activity monitoring is the measurement and interpretation of molecular signals to determine whether a biological signaling pathway is active, suppressed, or altered, making it valuable for understanding disease mechanisms and treatment responses. It typically combines readouts such as gene expression, protein abundance, phosphorylation, or downstream cellular effects with computational or experimental analysis to infer pathway status from complex biological data. In medicine, pathway activity monitoring supports disease classification, biomarker identification, therapeutic target evaluation, and patient-specific treatment strategies. By linking molecular changes to cellular functions and clinical outcomes, it helps researchers and clinicians assess disease progression and investigate how interventions influence biological systems.

2015

This report describes a method for measuring Drosophila larval activity using the TriKinetics Drosophila Activity Monitor. The device employs infrared beams to detect movements of up to 16 individual animals. Data can be analyzed to represent motion parameters including rates and the positions of the animals within the assay chambers.

Detecting SMAD Pathway Activation Using Dual Adenovirus Transduction

Source: Chen, H., et al. Live Cell Imaging of the TGF-β/Smad3 Signaling Pathway In Vitro and In Vivo Using an Adenovirus Reporter System. J. Vis. Exp. (2018).This video demonstrates a method to study SMAD pathway activation in breast cancer cells using dual adenoviral infection. One virus delivers a constitutively expressed GFP gene, while the other carries an RFP reporter under a SMAD-sensitive promoter. Upon TGF-beta stimulation, SMAD proteins are activated, forming a complex that induces RFP...

2018

Here we present a luciferase-based biosensor to quantify the kinase activity of large tumor suppressor (LATS)-a central kinase in the Hippo signaling pathway. This biosensor has diverse applications in basic and translational research aimed at investigating Hippo pathway regulators in vitro and in vivo.

Drosophila Activity Monitor (DAM): A Method to Measure Locomotor Activity in Flies

2023

This video describes the Drosophila activity monitor (DAM) system used to track locomotor activity. Researchers use activity data collected from the DAM to study circadian rhythms in fruit flies. The featured protocol clip shows how to load flies in the device and record activity data for circadian experiments.

Isothermal Microcalorimetry to Monitor Metabolic Activity in Bacteria

2025

Take serial dilutions of a bacterial culture and add aliquots into calorimetric vials containing growth medium.Load the sealed vials into the multi-well plate of an isothermal microcalorimeter to measure metabolic heat production. The device is held at a constant temperature to ensure the detected heat originates solely from bacterial metabolism. As bacteria metabolize nutrients and proliferate, they release heat that sensors detect in real time.In parallel, incubate identical vials in an...
