Patient Donor Selection

Patient donor selection is the clinical process of identifying a safe, suitable donor for a patient who requires donated blood, cells, tissue, or an organ. It combines assessment of donor health and eligibility with compatibility testing, including blood group, tissue markers, infectious disease screening, and other factors relevant to the planned procedure. Careful selection reduces the risk of immune-mediated complications, transmission of infection, and treatment failure while supporting ethical and equitable care. In transplantation and cellular therapies, structured donor evaluation helps clinicians balance biological compatibility, urgency, expected benefit, and procedural risk when choosing among potential donors.

Quantification of donor-derived cells is required to monitor engraftment after stem cell transplantation in patients with hemoglobinopathies. A combination of flow cytometry-based cell sorting, colony formation assay, and subsequent analysis of short tandem repeats may be used to assess the proliferation and differentiation of progenitors in the erythroid compartment.

Single Port Donor Nephrectomy

Single port laparoscopic surgery is changing the standard of care in surgical care like nothing since the laparoscopic technique was introduced 20 years ago. We present out technique of single port donor nephrectomy using the Gelpoint device. We have successfully performed this surgery in 100 patients.

Corneal Donor Tissue Preparation for Endothelial Keratoplasty

Endothelial corneal transplantation is a surgical technique for treatment of posterior corneal diseases. Mechanical microkeratome dissection to prepare tissue results in thinner, more symmetric grafts with less endothelial cell loss and improved outcomes. Dissections can be performed at the eye bank prior to corneal transplantation surgery.

Here we present a protocol describing oxygenated ex situ machine perfusion of donor liver grafts. This article contains a step by step protocol to procure and prepare the liver graft for machine perfusion, prepare the perfusion fluid, prime the perfusion machine and perform oxygenated normothermic machine perfusion of the liver graft.
