Peripheral Blood Stem Cells

Peripheral blood stem cells are blood-forming, or hematopoietic, stem cells found in the bloodstream and used to restore bone marrow function after intensive treatment. Because they normally reside mainly in bone marrow, medications such as granulocyte colony-stimulating factor can mobilize them into circulation, where leukapheresis separates and collects the stem-cell-rich fraction from the blood. The collected cells may be cryopreserved and infused into a patient after chemotherapy or radiation conditioning, then migrate to the bone marrow and regenerate blood and immune cells. This approach supports autologous and allogeneic transplantation for disorders including leukemia, lymphoma, multiple myeloma, and selected nonmalignant diseases.

This protocol describes a detailed method for efficient generation of integration-free iPSCs from human adult peripheral blood cells. With the use of four oriP/EBNA-based episomal vectors to express the reprogramming factors, KLF4, MYC, BCL-XL, or OCT4 and SOX2, thousands of iPSC colonies can be obtained from 1 mL of peripheral blood.

Here we show a simple and effective protocol for the generation of human iPSCs from 3-4 ml of peripheral blood using a single lentiviral reprogramming vector. Reprogramming of readily available blood cells promises to accelerate the utilization of iPSC technology by making it accessible to a broader research community.

Generation and Culture of Blood Outgrowth Endothelial Cells from Human Peripheral Blood

This protocol allows for the reliable generation and characterization of blood outgrowth endothelial cells (BOECs) from a small volume of adult peripheral blood. BOECs can be used as a surrogate for endothelial cells from patients with vascular disorders and as a substrate for the generation of induced pluripotent stem cells.
