Tumor Bioluminescence

Tumor bioluminescence is an optical imaging approach that uses light-producing reactions to visualize and measure cancer cells or tumors in living experimental models. Typically, tumor cells are engineered to express a luciferase enzyme; after administration of its substrate, such as luciferin, the enzyme catalyzes a reaction that emits photons, and sensitive cameras detect the signal through surrounding tissue. Signal intensity can indicate changes in tumor burden, distribution, or treatment response over time. In medicine, this method supports noninvasive monitoring of cancer progression, metastasis, and therapeutic efficacy while reducing the need for repeated tissue collection in preclinical research.

Bioluminescence imaging is a well-known tool for localizing tumors and metastases, but quantification of these images often requires complex calculations and particular instruments. We describe the easy-to-use luminoscore method, based on precise acquisition conditions, requiring no calculations, and enabling tumor burden and treatment response to be monitored in mouse models.

Establishing Intracranial Brain Tumor Xenografts With Subsequent Analysis of Tumor Growth and Response to Therapy using Bioluminescence Imaging

Luciferase-modified human brain tumor xenografts can be established intracranially in athymic mice, with subsequent monitoring of tumor growth and response to therapy using bioluminescence imaging. In combination with survival analysis, bioluminescence monitoring is an essential research tool for pre-clinical testing of therapies being considered for treating brain tumors.
