Apoptotic Stimulus Detection

Apoptotic stimulus detection is the identification of cellular signals that initiate apoptosis, a regulated form of programmed cell death. In neurons, stressors such as DNA damage, trophic-factor withdrawal, oxidative stress, or excitotoxic signaling can alter BCL-2 family activity, trigger mitochondrial outer-membrane permeabilization, and activate caspases; death-receptor pathways can also initiate caspase signaling. Researchers detect these stimuli and downstream events by measuring cytochrome c release, caspase activity, phosphatidylserine exposure, or DNA fragmentation. This approach helps distinguish regulated neuronal death from other forms of injury and supports research into neurodevelopment, neurodegenerative disease, and potential neuroprotective interventions.

To detect healthy cells in whole animals that contain low levels of caspase activity, the highly sensitive biosensor designated CaspaseTracker was generated for Drosophila. Caspase-dependent biosensor activity is detected in long-lived healthy cells throughout the internal organs of adult animals reared under optimized conditions in the absence of death stimuli.
