Cell Type Identification

Cell type identification is the process of distinguishing and classifying cells according to their molecular, structural, physiological, or functional characteristics. In neuroscience, researchers combine features such as morphology, gene-expression markers, protein profiles, electrophysiological behavior, and connectivity to determine whether cells belong to distinct neuronal or glial populations. Methods including immunohistochemistry, in situ hybridization, single-cell RNA sequencing, and patch-clamp recording provide complementary evidence, while computational analysis can integrate these measurements into cell-type classifications. Accurate identification supports studies of neural circuits, development, disease mechanisms, and therapeutic targets by linking cellular identity to brain function and pathology.

In this video, we describe a procedure for the expression of bacterial type III effectors in yeast and the identification of effector-induced growth inhibition phenotypes. Such phenotypes can be subsequently exploited to elucidate effector functions and targets.

Fiber Type Identification of Human Skeletal Muscle

This protocol demonstrates single-fiber isolation from freeze-dried human skeletal muscle and fiber-type classification according to Myosin heavy chain (MHC) isoform using the dot blotting technique. Identified MHC I and II fiber samples can then be further analyzed for fiber type-specific differences in protein expression using western blotting.
