Immunofluorescence Study

Immunofluorescence study is a microscopy-based technique that uses fluorescently labeled antibodies to detect and visualize specific molecules in cells or tissue. Samples are typically fixed and permeabilized, then exposed to primary antibodies that bind target proteins and fluorescent secondary antibodies or directly labeled probes; excitation light causes the fluorophores to emit detectable signals. In neuroscience, immunofluorescence helps map neuronal markers, receptors, synaptic proteins, and glial components within brain tissue or cultured cells. By revealing molecular localization, cell identity, and changes in protein distribution, the method supports research on neural development, connectivity, disease mechanisms, and responses to experimental treatments.

An In Vivo Immunofluorescence Localization Method to Study Antibody Biodistribution

2025

This video demonstrates an in vivo immunofluorescence localization (IVIL) technique to study the biodistribution of tumor-targeting antibodies in a murine model of breast cancer. Upon the intravenous injection of a tumor-specific antibody in the mouse, the tumor is surgically isolated, and the localization of the antibody within the tumor tissue is confirmed via ex vivo immunostaining.

A fluorescence microscope uses fluorescent chromophores called fluorochromes, which can absorb energy from a light source and then emit this energy as visible light. Fluorochromes include naturally fluorescent substances (such as chlorophylls) and fluorescent stains that are added to the specimen to create contrast. Dyes such as Texas red and FITC are examples of fluorochromes. Other examples include the nucleic acid dyes 4’,6’-diamidino-2-phenylindole (DAPI), and acridine orange. The...
