Late Neuronal Medium

Late neuronal medium is a specialized cell-culture formulation used to support neurons during later stages of differentiation, maturation, and maintenance in vitro. Its nutrients and signaling components help cells survive after initial neuronal induction while promoting mature neuronal features, including neurite development, synaptic organization, and functional activity; protocols typically introduce it after an earlier growth or differentiation medium. In neuroscience research, late neuronal medium supports the generation of more physiologically relevant neuronal cultures for studying development, connectivity, disease-associated phenotypes, and responses to experimental treatments. Consistent use of an appropriate formulation can improve culture stability and the reproducibility of downstream analyses.

This video demonstrates an assay for promoting the growth of neuron projections known as neurite outgrowth using the macrophage-conditioned medium. Co-culture neurons and macrophages with cAMP and harvest the macrophage-derived conditioned medium. Growing the neurons with the conditioned medium promotes neurite outgrowths. These outgrowths are fluorescently labeled and visualized using a fluorescence microscope.

Measuring left ventricular pressure (LV) in embryonic and neonatal mice is described. Pressure is measured by inserting a needle connected to a fluid-filled transducer into the LV under ultrasound guidance. Care must be taken to maintain normal cardiac function during the experimental protocol.
