Mouse Retinal Dissection

Mouse retinal dissection is a laboratory method for isolating the retina from the eye to study its structure, cells, and neural circuits in neuroscience. The procedure involves carefully removing the ocular tissues surrounding the retina, including the cornea, lens, and sclera, then separating the neural retina while preserving its layered organization for downstream analysis. Researchers use dissected retinas for histology, immunostaining, electrophysiology, gene expression studies, and imaging of retinal neurons and synaptic connections. This preparation supports investigations of visual processing, retinal development, neurodegeneration, and responses to injury or therapeutic interventions.

Xenopus laevis provides an ideal model system for studying cell fate specification and physiological function of individual retinal cells in primary cell culture. Here we present a technique for dissecting retinal tissues and generating primary cell cultures that are imaged for calcium activity and analyzed by in situ hybridization.

In this video, we will demonstrate how to isolate retinal stem cells from the ciliary epithelium of the mouse eye and grow them in culture to form clonal retinal spheres. The spheres that are isolated possess the cardinal properties of stem cells: self-renewal and multipotentiality.
