Nanoliter Injection

Nanoliter injection is a microscale delivery technique that introduces extremely small liquid volumes, typically in the nanoliter range, into cells, tissues, or experimental systems with high spatial and volumetric precision. In neuroscience, a fine glass micropipette is positioned in a target neuron or brain region, and controlled pressure or mechanical displacement expels a calibrated volume of solution while limiting local disturbance. Researchers use nanoliter injection to deliver tracers, pharmacological agents, genetic materials, or other reagents for studying neural circuits, cellular signaling, and brain function. Its precision supports localized experiments and helps reduce exposure of surrounding tissue.

Ice binding proteins (IBPs), also known as antifreeze proteins, inhibit ice growth and are a promising additive for use in the cryopreservation of tissues. The main tool used to investigate IBPs is the nanoliter osmometer. We developed a home-designed cooling stage mounted on an optical microscope and controlled using a custom-built LabVIEW routine. The nanoliter osmometer described here manipulated the sample temperature in an ultra-sensitive manner.

This protocol provides detailed guidance for the initial and continued generational allotransplantation of Drosophila tumors into the abdomen of adult hosts for studying various aspects of neoplasia. Using an autoinjector apparatus, researchers can achieve improved efficiency and tumor yields compared to those achieved by traditional, manual methods.

After neural tube formation, the neuroepithelium constricts and folds while the tube fills with embryonic cerebrospinal fluid (eCSF) to form the embryonic brain ventricles. We developed this ventricle injection technique to better visualize the fluid filled space in contrast to the neuroepithelial shape in a live embryo.

Modeling Neonatal Intraventricular Hemorrhage Through Intraventricular Injection of Hemoglobin

2025

This video demonstrates a method to generate a rat model of neonatal intraventricular hemorrhage. In this procedure, an anesthetized rat pup is injected with hemoglobin into the lateral ventricle of the brain. The hemoglobin causes oxidative stress and releases heme, simulating intraventricular hemorrhage. The resulting damage to brain tissues, driven by reactive oxygen species and inflammatory cytokines, leads to ventricular enlargement, a common consequence of intraventricular hemorrhage.
