Paraffin Oil Pool

A paraffin oil pool is a layer or reservoir of liquid paraffin placed over an exposed biological preparation to shield it from air and limit dehydration during microscopy or electrophysiology. In neuroscience, researchers form the pool around a dissected nervous system, often after removing surrounding tissue, so the oil covers the preparation while electrodes, probes, or imaging optics access selected structures. This arrangement helps maintain a stable environment for prolonged recordings and manipulation, particularly in small experimental organisms and semi-intact preparations. Paraffin oil pools therefore support studies of neural circuits, sensory physiology, synaptic function, and behavior by preserving tissue accessibility without enclosing the entire preparation in aqueous solution.

Detection of Rare Genomic Variants from Pooled Sequencing Using SPLINTER

Pooled DNA sequencing is a fast and cost-effective strategy to detect rare variants associated with complex phenotypes in large cohorts. Here we describe the computational analysis of pooled, next-generation sequencing of 32 cancer-related genes using the SPLINTER software package. This method is scalable, and applicable to any phenotype of interest.

FM dyes have been of invaluable help in the understanding of synaptic dynamics. FMs are normally followed under the fluorescent microscope during different stimulation conditions. However, photoconversion of FM dyes combined with electron microscopy allows the visualization of distinct synaptic vesicle pools, among other ultrastructure components, in synaptic boutons.
