Phenylephrine Injection

Phenylephrine injection is a parenteral medication that produces rapid vasoconstriction and is used to increase arterial blood pressure, making it relevant to studies of vascular and autonomic nervous system function. As a selective α1-adrenergic receptor agonist, it activates receptors on vascular smooth muscle, elevating systemic vascular resistance; the resulting pressure increase can also engage baroreflex pathways and slow heart rate. In neuroscience and neurophysiology, phenylephrine helps investigate blood pressure regulation, sympathetic control, baroreflex responses, and relationships between vascular tone and cerebral perfusion. Clinically, it is commonly used to manage acute hypotension, including episodes associated with anesthesia.

Source: Park, S. W., et al. Limbal Approach-Subretinal Injection of Viral Vectors for Gene Therapy in Mice Retinal Pigment Epithelium. J. Vis. Exp. (2015)The video describes the method for subretinal injection of viral vectors into the retinal pigment epithelium (RPE) in a mouse model. It details the procedure from anesthesia and eye preparation to precise lentiviral vector delivery, ensuring gene integration and expression in the RPE.

Subretinal injections are the most common technique for delivering large therapeutic agents such as proteins and viral vectors to photoreceptors and the retinal pigment epithelium. An alternative method in mice that successfully targets the subretinal space with minimal collateral damage and fast recovery times is described here.

After neural tube formation, the neuroepithelium constricts and folds while the tube fills with embryonic cerebrospinal fluid (eCSF) to form the embryonic brain ventricles. We developed this ventricle injection technique to better visualize the fluid filled space in contrast to the neuroepithelial shape in a live embryo.
