Stellate Morphology

Stellate morphology describes a star-shaped neuronal structure in which multiple processes, especially dendrites, radiate from the cell body. In neuroscience, this arrangement increases the membrane area available for synaptic contacts and enables the neuron to integrate inputs arriving from different directions; the number, branching pattern, and orientation of dendrites can shape signal processing within a circuit. Researchers use stellate morphology to classify neurons, relate cellular structure to electrophysiological function, and interpret connectivity in regions such as the cerebral cortex and cerebellum. Examining these features with microscopy, labeling, and quantitative morphometry supports studies of circuit organization, development, and neurological disease.

Here we describe a method for the isolation of hepatic stellate cells from mouse liver. For stellate cell purification, mouse livers are digested in situ and in vitro by pronase-collagenase treatment prior to density gradient centrifugation. This technique yields highly pure hepatic stellate cells.

Pathophysiological changes in the cardiac autonomic nervous system, especially in its sympathetic branch, contribute to the onset and maintenance of ventricular arrhythmias. In the present protocol, we show how to characterize murine stellate ganglia to improve the understanding of the underlying molecular and cellular processes.
